Cell Sorting

Sorter operation

Sorters are prepared 

Sample tubes

Suspension samples may be placed in 1.5ml Eppendorf tubes, 12 X 75mm tubes, 15ml or 50ml tubes.

Sample concentration

Recommended cell concentration is 1X106 to 5X106 per ml of buffer/medium.

Beckman Coulter Moflo Astrios can run at 10 000 events per second.

The nozzle size for Astrios is 100um.

Sample filtering

Samples must be filtered with 60um nylon mesh (provided in the facility) to remove clumps that might plug the instrument during sorting

Unfiltered samples will not be allowed to sort

For samples that tend to aggregate over time, you may add some DNase

Add 0.02mg/ml DNase type IIS to all cell preparation steps, including wash steps, to eliminate free DNA from broken cells that leads to aggregation.

Collection

Sorted cells can be collected into

Tubes: 1.5ml Eppendorf tubes, 12 X 75mm tubes, 15ml or 50ml tubes.

Plates: 96-well and 384-well plates

Culture media or buffer or serum may be used in the collection tubes/plates

It is recommended to spin the plates post sorting for 30-60s at 300G to help the cells to adhere and increase the number of colonies that will grow.

Training

All users will be trained by our facility staff and chargeable at $60/pax/equipment.

We also offer chargeable hands-on training programme which provides comprehensive coverage on the field. Please contact Wang Xiaoning (medwx@nus.edu.sg) to confirm the training slot.

In collaboration with industry partners, we also provide advanced training for operators of core facilities located in Southeast Asia.

Samples and some consumables are provided by the facility. The participants may try their own cell samples but please notify us in advance.

Policies

Service Charge and Scheduling Policy

The following rules apply to time scheduled on Flow Cytometry Facility instruments:

  • “No-shows” are charged for all of the scheduled time.
  • Appointments cancelled less than 24 hours in advance will be charged for the full time.
  • Appointments for the cell sorter cannot be scheduled less than 24 hours. 2 to 3 days in advance booking for cell sorting is recommended.

Invoice request

The Facility will generate quarterly invoices that are sent by mail to designated people. Invoices covering other ranges of time can also be generated upon request. If you require certified true copy of invoices, please send your request to medwx@nus.edu.sg .

Data Storage 

Data generated by BD LSRFortessa or Cytoflex LX are stored in a database local to the instrument PC. Once a user has completed data acquisition, files can be exported from the database in two formats, either as flow cytometry standard files (FCS) or as entire experiments. It is recommended that FCS files are exported to the instrument PC immediately after the completion of each run for off-line analysis.

Acceptable methods to transfer data:

  • Through facility thumbdrive (for Cytoflex LX and BD LSR Fortessa)

We encourage users to back up their data as soon as possible.

We are not responsible for any data loss in the case of computer breakdown or special case etc.
