Abstract:
Bats host a diverse array of paramyxoviruses including multiple species with documented spillovers into human populations. Receptor usage not only determines the entry of a virus into a new host and thus the potential for successful spillover events, but also the tissue tropism and pathogenic sequelae of the resulting virus infection. Here, we evaluate the role of receptor usage in the zoonotic potential and pathogenesis of novel bat-borne paramyxoviruses related to known human pathogens such as Nipah virus, measles virus and mumps virus. Some of these viruses are known only from metagenomic surveys. We show how we developed novel systems and animal models to assess the pathogenesis of these viruses, and their susceptibility to next generation direct-acting antivirals.
